Indications for Use
Human oocyte and embryo washing/manipulation and fertilization by ICSI.
Storage and Shelf Life
Store at 2-8°C and protected from light. Ten (10) weeks from the date of manufacture.
Disposal Consideration
Treat or dispose of waste material in accordance with all local state/provincial, and national requirements. Dispose with laboratory waste.
Composition
A HEPES-buffered medium is required when oocytes and embryos are handled outside of a CO2 incubator. Sodium Chloride, Potassium Chloride, Calcium Chloride, Potassium Phosphate, Magnesium Sulfate, Sodium Bicarbonate, Glucose, Lactate Na Salt, Sodium Pyruvate, Glycine, L-Alanine, L-Arginine HCl, L-Asparagine, L-Aspartic Acid, L-Cystine, L-Glutamic Acid, Glycyl-Glutamine, L-Histidine, L-Isoleucine, L-Leucine, L-Lysine HCl, L-Methionine, L-Phenylalanine, L-Proline, L-Serine, L-Threonine, L-Tryptophan, L-Tyrosine, L-Valine, EDTA, Phenol Red, HEPES, Human Serum Albumin* (5 mg/ml), Gentamicin Sulfate* (10 µg/ml) *from therapeutic-grade source material

INSTRUCTIONS FOR USE
The procedures described below have been found to be effective for human oocyte and embryo washing/manipulation and fertilization by ICSI. Every laboratory must define and optimize its own procedures.
1. For oocyte and embryo washing and manipulation, prepare culture dishes containing 25-100 μl droplets under oil, or in larger volumes (0.5-1.0 ml) of global® total® LP w/ HEPES, according to general laboratory practice.
2. Before introducing the oocytes or embryos, warm the culture dishes for sufficient time to ensure temperature equilibration.
3. For washing, pass the oocytes or embryos through several droplets of global® total® LP w/ HEPES.
4. For ICSI, prepare 5-10 small droplets (5-10 µl) of global® total® LP w/ HEPES in the ICSI dish, next to the PVP droplets and cover them with oil, according to general laboratory practice. Transfer the oocytes to the respective droplets just before the ICSI procedure.
